In Vitro Laboratory Use

Reconstitution and storage conditions:

  • Dissolve in sterile water or phosphate buffer (recommended pH 6.0–7.4); working concentration per analytical protocol
  • Storage of unreconstituted powder: −20 °C, dry location, protected from light
  • After reconstitution: aliquots in microtubes, single thaw, short-term storage at 2–8 °C

Typical analytical equipment:

  • HPLC column dedicated to peptide analysis (e.g., C18, 150–250 mm)
  • HPLC system with detector (UV 220 nm, FLD or MS — depending on protocol)
  • Autosampler vials, laboratory test tubes, precision pipettes
  • Analytical balance with ≥0.1 mg accuracy for sample weighing

All working parameters should be selected according to the research laboratory’s internal protocol. Product intended exclusively for in vitro and analytical use; not for human or animal use.

Molecular Mechanism

PT-141 is a cyclic heptapeptide and synthetic analog of alpha-melanotropin (alpha-MSH). Structurally, it is an active metabolite of Melanotan II, differing by the absence of an amide group at the C-terminus. PT-141 acts as a non-selective melanocortin receptor agonist, with affinity for MC1R, MC3R, MC4R, and MC5R subtypes, while showing no activity at the MC2R (ACTH) receptor.

In in vitro and animal model studies, activation of MC4R in the central nervous system was shown to modulate signaling pathways related to neuroendocrine functions. The cyclic lactam structure confers increased proteolytic stability compared to linear melanocortin analogs.

PT-141 vs Melanotan II

PT-141 and Melanotan II are structurally related melanocortin peptides with different pharmacological profiles. PT-141 is the active metabolite of Melanotan II, formed by removal of the C-terminal amide group.

Stability and Storage

In lyophilized form, store PT-141 at -20°C for long-term stability. After reconstitution in sterile water, the solution remains stable for up to 30 days at 2-8°C. The cyclic lactam structure provides higher resistance to enzymatic degradation compared to linear melanocortin peptides.

Related Research Materials

PT-141 belongs to the melanocortin peptide family, which also includes Melanotan II and other alpha-MSH analogs. The cyclic lactam structure of PT-141 exemplifies a modification that enhances proteolytic stability of linear peptides.

Frequently Asked Questions

PT-141 (Bremelanotide) is a synthetic cyclic heptapeptide and analog of alpha-melanocyte-stimulating hormone (alpha-MSH). It acts as an agonist at melanocortin receptors MC1R, MC3R, MC4R, and MC5R, with preference for MC4R pathways in the CNS.

PT-141 is a metabolite of Melanotan II with greater selectivity for MC3R/MC4R over MC1R. Melanotan II is a broader melanocortin agonist affecting MC1R/MC3R/MC4R/MC5R. Both are cyclic heptapeptides derived from alpha-MSH.

Store lyophilized PT-141 at -20°C to -80°C, protected from light. After reconstitution, store at 2-8°C and use within 30 days.

PT-141 is available as a 10 mg lyophilized vial.

Every batch undergoes RP-HPLC analysis (≥98%) and mass spectrometry verification. Certificate of Analysis included.
