PT-141 is a cyclic heptapeptide and synthetic analog of alpha-melanotropin (alpha-MSH). Structurally, it is an active metabolite of Melanotan II, differing by the absence of an amide group at the C-terminus. PT-141 acts as a non-selective melanocortin receptor agonist, with affinity for MC1R, MC3R, MC4R, and MC5R subtypes, while showing no activity at the MC2R (ACTH) receptor.
In in vitro and animal model studies, activation of MC4R in the central nervous system was shown to modulate signaling pathways related to neuroendocrine functions. The cyclic lactam structure confers increased proteolytic stability compared to linear melanocortin analogs.
PT-141 and Melanotan II are structurally related melanocortin peptides with different pharmacological profiles. PT-141 is the active metabolite of Melanotan II, formed by removal of the C-terminal amide group.
